While in MyDVD creating a dvd project, is it possible to mute or lower the audio from the video while playing an added audio track over it at a normal volume?

Yes, I assume you mean the 'movie' and not the menu background:
Select the movie first
Click on EDIT MOVIE
Right click on the video and select edit>Native audio. There you can adjst the volume of the native audio in the video file.
Click OK to get back to edit screen
Then add your audio file to music track.
