The program will no longer launch after converting my system from FAT32 to NTFS. It will show the loading assets window for about 2 seconds, and then nothing happens. Is there a way to fix this? My reason for converting to NTFS was to transfer larger files between my computers.

you have to do a clean install.  uninstall the software and then go into the registries and clear out DVDit Pro HD from the Roxio area under HKEY_CURRENT_USER -> Software and under HKEY_LOCAL_MACHINE -> Software.  pretty good instructions here:
