Can Roxio Label Creator be configured to print to Epson Stylus Photo 2100?  As far as I know it was one of the first direct to disc printing models Epson released so I was expecting to find it there.

This seems to be an area not covered in any way at all by the FAQ or Roxann database.

When you go to print, does the top most box recognize that you have that printer attached?  After you select print to CD/DVD,  you indicate that your printer is not listed.  Is that correct?

You may want to look at the other Epson printers listed to see if the position of the disc in the tray is the same.  Try printing on a thin cardboard blank and then adjusting the offset if you are close.  Let us know how you make out.
Regardless of what I say about computer maintenance, there is no need to defrag a solid state hard drive.
