Have purchased download edition. Have downloaded Roxio Creator 2010 from Order Completed page. Do I also download, from same page, Roxio Creator 2010 Content?

Yes, you download anything that you can see available there to download.
The Program file is the first to install. That installs all the working programs and a few themes and menus and things. The Content is next, and that installs extra themes, menus, and pretty bits.
Be sure to burn your download files onto a disc as backup, because you'll need them again if you ever need to reinstall.
